Poly-L-Lysine (PLL) is a polycation formed by the polymerization of lysine monomers. It has two isomers, left-handed and right-handed. The left-handed one, PLL, is commonly used. PLL with a relative molecular weight of 70,000 to 300,000 can be used as a growth matrix for cell culture, which can promote cell adhesion and adherence to the wall. It is a commonly used cell adhesive in cell culture. The common molecular weights of poly-L-lysine (PLL) are 70,000 to 150,000, 150,000 to 300,000 and >300,000. The larger the molecular weight, the stronger the adhesion, but it is relatively difficult to completely dissolve. Poly-L-lysine with a molecular weight of 150,000 to 300,000 is generally used in cell culture.
Instructions for use (cell culture)
1. Dilution: dilute the poly-L-lysine solution to the working concentration (0.1 mg/mL) with sterile purified water or PBS solution at a ratio of 9:1. The diluted poly-L-lysine solution is effective for at least 3 months when stored at 2℃~8℃.
2. Coating: add the poly-L-lysine solution of the working concentration to the culture vessel. The amount added depends on the size of the vessel. Generally speaking, it can wet or cover the growth surface of the culture vessel. Let it stand at 37℃ for 2h~4h or at room temperature overnight. Operate aseptically.
3. Washing: remove poly-L-lysine, wash the culture vessel several times with sterile purified water or PBS solution, and use it after sterile air drying on the clean bench.
4. Wash the culture vessel once with culture medium before inoculating cells, and then inoculate the cells into the coated culture vessel.
5. Observe the growth status of the cells under the microscope the next day.
Precautions
1. This product is sterilized by filtration. When using this product, you should pay attention to aseptic operation to avoid contamination.
2. If the poly-L-lysine solution is turbid or contaminated after use, it should be discarded.
3. This product is only used for scientific research or further research, not for diagnosis and treatment.
