RPMI 1640 was developed by Moore et al. in 1967 at the Roswell Park Memorial Institute (RPMI) in Farrow, New York, USA. RPMI is a type of cell culture medium developed by the institute, and 1640 is the code name of the culture medium. RPMI 1640 is an improved McCoy's 5A medium that uses a bicarbonate buffer system. Unlike most mammalian cell culture media, its typical pH 8 formula is different. RPMI 1640 medium was originally designed for lymphocyte culture. It is now widely used in the culture of various normal cells and cancer cells, especially suspension cells. It is one of the most widely used culture media.
HEPES is an excellent biological buffer that is non-toxic to cells. The culture medium with HEPES added can maintain a constant pH range for a long time, which can effectively prevent the adverse effects of changes in the pH of the culture medium on cell growth.
Penicillin-streptomycin mixture is also commonly known as "double antibiotic". It is the most commonly used antibiotic in in vitro culture to prevent microbial contamination. Among them, penicillin can interfere with the synthesis of bacterial cell walls and is particularly effective against Gram-positive bacteria; while streptomycin can bind to the 30S subunit of bacterial ribosomes and inhibit the synthesis of bacterial proteins. It is effective against both Gram-negative and Gram-positive bacteria, but is particularly effective against Gram-negative bacteria. The combined use of penicillin and streptomycin can prevent most bacterial contamination.
This product contains a variety of ingredients such as amino acids, vitamins, inorganic salts, etc. required for various types of cell culture; but it does not contain proteins, lipids or any growth factors; therefore, this product needs to be used with serum or serum-free additives.
