RPMI (Roswell Park Memorial Institute) 1640 medium was originally developed for the culture of human leukemia cells in suspension and monolayer. RPMI 1640 medium has now been found to be suitable for a variety of mammalian cells, including HeLa, Jurkat, MCF-7, PC12, PBMC, astrocytes, and cancer cells. RPMI 1640 medium is unique from other media because it contains the reducing agent glutathione and high concentrations of vitamins. RPMI 1640 medium contains biotin, vitamin B12, and PABA, which are not found in MEM or DMEM, and is also very high in vitamins inositol and choline. RPMI 1640 medium does not contain proteins, lipids, or growth factors, so it requires supplementation with other reagents, usually 10% fetal bovine serum. RPMI 1640 medium uses a sodium bicarbonate buffer system (2.0 g/L) and requires a 5-10% carbon dioxide environment to maintain physiological pH.
